蛋白质表面修饰及与DNA的相互作用研究

摘要:蛋白质是构成人体组织与器官的主要物质,DNA是遗传信息的载体,蛋白质和DNA对生命体都是极其重要的,它们在生物体内扮演着不同但至关重要的角色。人体内蛋白质的种类数不胜数,不同的蛋白质因为结构不同自然有着不同的功能。但是理论上大部分的蛋白质都可以通过一定的化学方法与DNA共轭交联,得到的交联体的用途也各不相同,因此可知,蛋白质-DNA交联体具有广阔的发展前景。近年来,DNA-蛋白质相互作用的研究越来越为人瞩目,已经成为21世纪生命科学研究的主要课题之一。本文紧跟时代前沿,采用双异官能团交联试剂Sulfo-SMCC(中文名为4-(N-马来酰亚胺甲基)环己烷-1-羧酸磺酸基琥珀酰亚胺酯钠盐,sSMCC)先与巯基修饰的DNA单链共价结合,再与带有氨基的血红蛋白反应,从而实现了蛋白质和DNA的交联。之后再用DNA的互补链与其完成自组装,形成一个带有一个或多个蛋白质的DNA双链复合体。之后再用蛋白质的聚丙烯酰胺凝胶电泳、DNA的琼脂糖凝胶电泳、紫外吸收光谱等方式对复合体进行表征。表征结果却并不能说明复合物是否已经被合成成功,仍需要继续研究。但我们预测该复合体是一种具有生物活性的结构新颖的超分子物质,在疾病治疗及生物分子自组装研究领域存在广阔的发展前景。

Surface modification of proteins and the study of Protein-DNA interactions

Abstract:Proteins are the main material of the tissues and organs of the human body. DNA is the carrier of human genetic information. So, DNA and proteins are extremely important for life , they play a different but important role in human body. And there is an almost endless variety of proteins, the functions of those proteins are also not the same because of their different structures. But the most of proteins can interact covalently with DNA using a cross-linker by a chemical method in the theory. And the complex can be used in different ways, which indicates the proteins-DNA conjugate has a broad prospect of development. In recent years, the research of DNA-Protein interaction attracts more and more attention. It has become one of the main research topics of bioscience in the twenty-first Century. Following the forefront of the times, I use heterobifunctional cross-linking reagent Sulfo-SMCC (Chinese named 4- (N- maleimide methyl cyclohexane carboxylic acid sulfonic acid) -1- hydroxysuccinimide ester sodium salt, sSMCC) combine covalently with thiol-modified DNA single strand and then react with the amino groups of hemoglobin for realizing the cross-linking of Proteins and DNA. After DNA and complementary strand DNA completing self-assembly , the complex that consists of a protein with two DNA double chains has been synthesised successfully. After that, we use the protein gel electrophophoresis of polyacrylamide, the DNA gel electrophoresis of agarose and UV absorption spectroscopy or other methods to detect the complex. But the results do not indicate whether the complex has been successfully synthesized, and still need to study. We predict that this complex is a kind of` novel supramolecular material with biological activity. And the complex has broad prospects for development in the treatment of disease and biological molecular self-assembly field .
